INTERLEUKIN-4 & INTERFERON-gamma DETERMINATION PROGRAM DOCUMENT
The IL4IFNG program is written in GW-BASIC and it can run without math co-processor.
IL4-IFN is desigen to estimate the concentration of unknown samples of
IL-4 and IFN-$\gamma$ by utilizing the known standard data of these lymphokines.
This program is entirely menu driven, follow the menu to run the program.
The main menu provides the following five options.
ENTER AND SAVE THE STANDARD DATA:
This option allow the users to enter and save the standard data in
an ASCII file. The user should enter the new filename when the program
ask the file name, for avoiding the overwrite on the old file. The user
should enter the IL-4 and IFN-$\gamma$ concentrations and corresponding IgG1
and IgG2a secretion respectively of
standard samples (known samples) determined by ELISA method in
B cells. The program save the data of standard samples in filename
entered by user with extension IL4 and IFN for IL-4 and IFN-$\gamma$
respectively, called the standard data file.
This standard data file can be used for the drawing
standard curve and for determination the IL-4 concentration.
DRAW THE STANDARD CURVE FROM DATA FILE:
This option allow the user to draw a calibration curve by using standard
data file.The user should enter the old filename
(previously saved standard data file) from directory displayed when the
program ask the file name. The program first calculate the usable range
(initial portion of the curve with positive slope) of standard data and then
fit the standard data in usable range using the least square curve fitting
method by draw a calibration curve.
You may get a hardcopy with the PrintScrn key to invoke the printer.
However, some printer does not support the graph mode. Only the text can
be printed out. Under such circumstances, you need to refer to your printer
for graphic mode support, or you have to use other screen dump program
to print the graph.
CALCULATION OF IL-4 & IFN-gamma CONCENTRATION:
This option allow the user to determine the IL-4 \& IFN-$\gamma$ concentration
of unknown samples by using there standard data files. The user should enter
the old filename (previously saved standard data file) from directory
displayed when the program ask the file name. The program uses the
least square curve-fitting method by using standard data of file and
calculate the IL-4 \& IFN-$\gamma$ concentration of unknown samples
corresponding to there IgG1 and IgG2a secretion respectively.
MODIFY THE STANDARD DATA:
This option allow the user to edit the standard data of data file. The
user should enter the old filename (previously saved standard data file)
from directory displayed when the program ask the file name. The program
provide the three option to user 1) to add the more data to file. 2) to
modify any data of the file 3) to remove the data from file. The program
also sort the data and overwrite the new data on old data in same data file.
PRINT THE DATA
This option allow the user to print the data of data file on printer. The
user should enter the old filename (previously saved standard data file)
from directory displayed when the program ask the file name. The program
print the IL-4 \& IFN-$\gamma$ concentrations and corresponding peak currents
of standard samples respectively.
PLOT STANDARD CURVE
The standard curve of IL-4 \& IFN-$\gamma$ can be plot by using HPGL plotter
or using HPGL emulsion of the plotter.
