| Cohort | C57BL/6 mice and B6.129S6-Cd4tm1Knw/J (CD4 knockout (KO)) mice were obtained from The Jackson Laboratory or the Na- tional Cancer Institute. B6.129P2-Cxcr6tm1Litt/J (Cxcr6 eGFP/eGFP , hereafter referred to as Cxcr6 KO) mice, have an in frame substitution of eGFP for Cxcr6, which allows in vivo tracing of Cxcr6 promoter expression and regulation, were kindly provided by Dr Joshua Farber. |

| Disease Introduction Mechanism | Pneumocystis is an opportunistic fungus that causes disease in immunocompromised individuals but does not cause clinical pa- thology in healthy individuals. Serological and pathologic studies suggest that most humans have been infected with Pneu- mocystis by 2e4 years of age. Early in the HIV/AIDS epidemic, Pneumocystis pneumonia (PCP) was one of the most common and lethal opportunistic infections seen in people with HIV (PWH) and correlated with low CD4þ T cell counts (<200 cells/ml), highlighting the critical importance of these cells in immunity to PCP. |
